Abstract: Purpose: This study developed and characterized novel silk fibroin nanoparticles (FNPs) functionalized with poly(vinyl pyrrolidone) (PVP) to deliver and controlled release the active ingredient diclofenac (DC). Methods: FNPs were prepared by the co-condensation method with ethanol (99.5%) as a precipitation agent, at a fibroin (SF)/PVP (0%, 1%, or 3%) to ethanol ratio of 1:7. The particles were loaded with DC at different initial amounts by both the co-condensation and adsorption methods. The products were physicochemically characterized in terms of drug entrapment efficiency, chemical interactions, particles sizes, and in vitro release in simulated digestive condition. Results: The FT-IR analysis of particles showed characteristics signals of SF, PVP, and DC, indicating that DC was successfully loaded into nanoparticle systems, with limited interferences from the formulating processes. The FNPs formulations had particle sizes from 51.67 to 2777.33 nm, dependent on the compositions. The DC entrapment efficiency was approximately 20%. The process of DC release from the particles was hindered in HCl pH 1.2 and accelerated in phosphate buffer pH 6.8, suggesting that the particles could protect and control the drug release in the digestive tract. Conclusion: The PVP functionalized FNPs incorporating DC was successfully developed and characterized, with optimal properties for oral delivery. The data shed more insights on further research on FNPs for oral drug delivery.
